method:-
In
mechanical method cells are kept in a suitable plasmolyticum and cut with a
fine knife so that protoplast are released from cells cut through the cell wall
,when the tissue is again
deplasmolysed.This method is suitable for isolation of protoplast vacuolated
cells.e.g of onion roots,radish roots
etc.
(B)
Enzymatics Method :- The enzyme method is almost used for isolation of protoplast.Sometimes
for mechanical and enzymatic method are combined .where cells are separated to
mechanically and later used for isolation of protoplast through enzymatic
treatment.
The protoplast can be
isolated from of variety of including roots,leaves in shoot culture .
However the
most commonly used organs are leaves which can be isolation of protoplast using
to following steps.
(i)Sterilization of leaves.
(ii)Peeling of the epidermis.
(iii)Enzymatic treatment and
(iv)Isolation and cleaning of protoplast.
